双波长比吸光度法测定甾类化合物HMPDD脱氢转化率

摘  要:建立了一种简便、快速的双波长比吸光度法,用于测定甾类化合物HMPDD和相应脱氢产物HMPTD组成的混合物中的HMPTD质量分数,其质量分数与HMPDD和HMPTD混合物的比吸光度(A267/A253)线性相关,回归方程为Y=3.05X-0.739,相关系数0.999 5.分别采用双波长比吸光度法和HPLC法对不同HMPTD质量分数的微生物脱氢转化样品进行测定,两者的相对误差小于2%,表明建立的双波长比吸光度法可用于HMPDD微生物脱氢反应过程的中间检测分析.A rapid and simple method for quantitative determination of 17crhydroxy-16]3-methylpregna-1,4,9, (11)-triene-3,20-dione (HMPTD) was investigated for mixed samples of HMPDD and HMPTD with an ahsorhance-ratio method of dual wavelength spectrophotometry. It was found that the mass fraction HMPTD related linearly to the ahsorhance ratio A267/A253. They can he correlated by using the following linear equation Y=3.05X-0. 739 with a coefficient of determination of 0. 9995. The method of dual wavelength spectrophotometry was used to analyze the samples obtained by microbial transformation in comparison with the HPLC method. The relative error was below 2%. This means that the proposed method was suitable for the measurement of the HMPDD conversion ratio for microbial dehydrogenation.
