摘  要:目的:研究阿魏酸钠(SF)对局灶性脑缺血再灌注后促进神经功能恢复和血管生成的作用。方法:制作大鼠局灶性脑缺血再灌注模型,随机分为治疗组和对照组,观察术后不同时间点大鼠神经功能、脑梗死体积、脑含水量的变化。用异硫氰酸荧光素(FITC)标记的右旋糖苷标记血浆,激光扫描共聚焦显微镜下观察缺血后7d脑血浆灌注量,脑血管形态和脑微血管直径的变化。结果:阿魏酸钠治疗组在术后2d、7d、14d的神经功能恢复均优于对照组。两组脑梗死体积均在术后第7d达到高峰,SF组梗死体积在各个时间点上均小于对照组,差异有显著性意义(P<0.05)。SF组脑含水量在术后2d高于缺血对照组和假手术组,随后逐渐下降,到术后14d低于缺血对照组和假手术组,差异有显著性意义(P<0.05)。术后7d,SF组脑血浆灌注量明显高于对照组,直径<3μm的血管约占47%,新生血管增多,呈不规则迂曲形态。结论:SF能促进脑缺血后神经功能的恢复,减小梗死体积,减轻脑水肿,其机制可能与促进血管生成的作用有关。Objective:To study the effects of sodium ferulate on neurological function and angiogenesis after reperfusion of cerebral ischemic in rats.Method:The change of neurological function, infarct volume and brain water content were measured at different time point after cerebral ischemia. The vessel diameters, morphology and volumes of plasma perfusion were analyzed on cerebral microvessels perfused with FITC-dextran 7 days after ischemia using LSCM.Result:The SF-treated group recovered better than saline-treated group at 2, 7, 14 days after surgery. The brain infarct volume was peaked on day 7 in both groups. The infract volumes of SF-treated group were significantly small compared with saline-treated group in each time point(P〈0.05). The brain water contents of SF-treated group were much higher than control group at 2 day, then diminished gradually, and were significantly lower in 14 day than saline-treated group and sham group(P〈0.05). The volumes plasma perfusion of SF-treated group were significantly increased compared with the control group. 47% of plasma perfused vessels in the penumbra had diameters within 3μm. The newly grown vessels were increased and had an irregular pattern of tortuosity.Conclusion: Sodium ferulate can improve functional neurological recovery, diminish infarct volumes and brain water contents. The possible mechanism is that the SF can enhance angiogenesis.
