摘  要:本实验用PCR扩增了来自世界各地的P.decipiens复合种共6个姊妹种的线粒体NADH脱氢酶亚单位Ⅳ基因(nad4),并进行了克隆、测序和序列分析。结果显示,PdOe与Pd为同一种,nad4基因序列种内的变异为0.7%,种间差异为2.4~13.2%。种间差异远大于种内变异,能提供准确的遗传标记。可用于P.decipiens复合种姊妹种的鉴定。这些结果有助于P.decipiens复合种的生活史、传播途径、流行病学、种群遗传学的研究和其所引起的相关疾病的诊断。The present study examined the nucleotide variation in a portion of the mitochondrial (mt) NADH dehydrogenase subunit 4 (nad4) between members of the Pseudoterranova decipiens(s, l. ) complex using a PCR approach. The objective was to assess whether this mtDNA gene sequence can provide genetic markers for the identification of mombers of the P. decipiens complex. The results showed that sequence differences in the nad4 gene between species were 2. 4~13. 2%, and that sequence variation within species was just 0.7%, indicating that had4 gene sequence provides genetic markers for the identification of 6 numbers of the P. decipiens complex. These findings should have important implications for studying life cycles, transmission patterns, epidemiology and population genetics of members of P. decipines, and for the diagnosis of diseases they cause.
