肝气郁结模型大鼠T细胞免疫功能的改变及柴胡疏肝散的治疗作用

摘　　要：目的:研究肝气郁结证(肝郁证)模型大鼠T淋巴细胞免疫功能的改变及柴胡疏肝散的治疗作用。方法:体外培养模型大鼠T淋巴细胞,分别应用MTT法、ELISA法、RT-PCR法检测肝气郁结证模型大鼠以及运用柴胡疏肝散、四君子汤治疗后T细胞的增殖能力、细胞因子IFNγ-、IL-4的含量及mRNA的表达。结果:与正常组比较,肝郁证模型大鼠T淋巴细胞的增殖能力下降,其增殖指数明显降低(P<0.05);IL-4细胞因子的含量及mRNA表达水平明显降低(P<0.05),IFNγ-细胞因子的含量及mRNA表达水平则明显升高(P<0.05),Th细胞向Th1向偏移。用柴胡疏肝散治疗后,与正常组相比较,各项免疫指标基本恢复了正常水平(均P>0.05),Th1、Th2趋于平衡状态。结论:肝气郁结证模型大鼠T淋巴细胞免疫功能紊乱,主要表现在增殖能力下降,Th1/Th2细胞因子处于失衡状态,Th细胞向Th1偏移;柴胡疏肝散可以纠正其T淋巴细胞免疫功能的紊乱状态。Objective： To investigate Changes of T cellular immune function in the rats with liver-qi stagnation syndrome and effect of powder of ChaiHnShuGan on it. Methods： T cells of models were cultured in vitro, then T lymphocyte proliferative ability, the content and mRNA of IL-4 and IFN-γ were detected by MTT, ELISA and RT-PCR respectively, and decoction of ShiJunZi was given. Results： Compared to normal group, the T lymphocyte proliferative ability was decrease, and the content and mRNA of IL-4 were decreased, while the content and mRNA of IFN-γ was increased （all P 〈 0.05）, Th cell shift towards Thl in the T cells of the model＇ s.Treated with powder of ChaiHuShuGan for mthods, could regulate immune function and convert the imbalant state to normal state. Compared with normal group, all index reached to normal levels （ P 〉 0.05 ）, and convert the imbalant state of Th1/Th2 and make it incling to normal state. Conclusions： T cellular immune function in the rats with liver -qi stagnation syndrome was disorder, it seems to be that the T llymphocyte proliferative ability decreased and imbalance in the Thl and Th2 arose, Th cell shifted toward Thl ; powder of ChaiHuShuGan could regulate immune function and convert the imbalant state to normal state.
