摘  要:目的:探讨利用糖原合成酶激酶3的抑制剂氯化锂对葡萄糖6磷酸酶基因表达的影响。方法:将原代培养的大鼠肝细胞在葡萄糖和不同浓度氯化锂的培养液中培养4h后提取RNA,以半定量RT-PCR方法检测细胞葡萄糖-6-磷酸酶催化亚基(G6PC)和6磷酸葡萄糖转运亚基(G6PT)的mRNA水平。结果:氯化锂(10mM、20mM)与高糖培养液联合培养肝细胞4h后,肝细胞葡萄糖-6-磷酸酶的催化亚基和转运亚基的mRNA的丰度均下降(P<0.05)。结论:锂盐可能能够有效的抑制葡萄糖-6-磷酸酶的表达起到胰岛素增敏的作用。对葡萄糖-6-磷酸酶进行干预能够有效的改善糖尿患者的高血糖状态。Objective: To study the effect of LiC1 on the expression of rat hepatocype glucose-6- phosphatase. Methods: The primary hepatocyte was cultured in different concentrations of glucose and LiC1. Total RNA was extracted after 4 hours incubated with glucose or LiC1. The mRNA of glucose-6- phosphatase catalytic (G6PC) gene and the putative glucose-6-phosphate translocase (G6PT) gene were measured by semiquantitative RT-PCR. Results: The rat hepatocyte cultured with the medium containing LiCl-the inhibitor of GSK and Glu(25mM) for 4 hours, both the mRNA level of P36 and P46 decreased significantly (P〈0.05). Conclusion: The inhibitor of glycogen synthase kinase LiC1 can both decrease the mRNA level of P36 and P46.The hyperglycemia of diabetes can be amelioratea by intervening the experssion of glucose-6-phosphatase.
