摘  要:研究了卡那霉素和6种头孢霉素(头孢曲松钠、头孢噻肟钠、头孢哌酮钠、头孢唑啉钠,头孢拉定和头孢他啶)对烟草叶片分化及对根癌农杆菌LBA4404和烟草转化植株的抑制效果。结果发现:不同烟草品种对卡那霉素的耐受性差异较大,品种CV为受体材料时,叶分化和茎段根再生的适宜浓度为50mg/L。品种T12为受体材料时,叶分化的适宜浓度为100mg/L,而茎段根再生则为200mg/L;头孢曲松钠和头孢噻肟钠对根癌农杆菌LBA4404的抑制效果好,并可使继代3次(60d)的烟草转化植株脱菌;不同浓度的6种头孢类抗生素对烟草叶片的分化影响不大。从头孢类抗生素对农杆菌和烟草转化植株的抑制作用、外植体的分化影响来考虑,在用农杆菌LBA4404作为介导菌株时,可选用200mg/L的头孢曲松钠或头孢噻肟钠来抑菌。Effects of Kanamycin and 6 cephalosporins, i.e. Ceftriaxone sodium, Cefotaxime sodium, Cefoperzanoe sodium, Cefazolin sodium, Cefradine, Ceftazidime, on differentiation of tobacco leaf and inhibition of Agrobacterium tumefaciens LBA4404 and tobacco transgenic plant were analyzed. The results showed that different tobacco varieties' tolerance toward Kanamycin were various. For the variety CV, the proper selective concentration of both differentiation of tobacco leaf and regeneration of roots were 50 mg/L, while for the variety T12, the proper selective concentration of differentiation of tobacco leaf was 100 mg/L and the regeneration of roots was 200 mg/L. Ceftriaxone sodium and Cefotaxime sodium showed good effects on inhibiting Agrobacterium tumefaciens LBA4404 and could eliminate the tobacco transgenic plant' s bacterial cells after three generations(60 d). There was a little effect of the 6 cephalosporins in different concentrations on differentiation of tobacco leaf. Considering the inhibition of cephalosporins on Agrobacterium tumefaciens LBA4404 and tobacco transgenic plant and the effects of cephalosporins on differentiation of explant, it is suggested that 200 mg/L of Ceftriaxone sodium or Cefotaxime sodium should be used to inhibit bacterial cells while Agrobacterium tumefaciens LBA4404 was the mediator.
