摘  要:目的探讨放射性碘标记抗IGF1R单克隆抗体1H7的实验条件及其性质。方法131I-1H7采用Iodogen法标记,Sephadex G-50层析柱分离纯化;纸层析测标记物放射化学纯度(RCP),分别置于室温、4℃、-20℃,于第8 d、16 d、24 d测定各自RCP,评价其体外稳定性,以竞争结合法鉴定其生物学性质。结果131I标记率为64.6%,131I-1H7放射性比活度5.48 MBq/μg,RCP 96.42%,即使在室温放置24 d后RCP仍达91.8%;131I-1H7与HepG2细胞结合的亲和常数K值为1.81×1011L/mol。结论Iodogen法131I标记1H7,所得产物放射化学纯度及放射性比活度高,体外稳定,生物活性保持完好,为进一步研究奠定了基础。Objective To explore how to label 1 H7 ( monoclonal antibody against type 1 receptor of insulin-like growth factor) with ^131 I and appraise the quality of the product. Methods 1 H7 was labeled with ,^131 I by Iodogen method, then purified with Sephadex G-50 column. The radiochemical purity (RCP) of the product was determined by paper chromatography. After stored at room temperature, 4 ℃, - 20 ℃ for 8, 16, 24 d respectively, the product was detected for RCP to evaluate its stability. Results The labeling rate was 64.6%, the specific activity and RCP of ^ 131I-1 H7 was 5.48MBq/μg and 96.42% respectively. Even the product stored at room temperature for 3 half-lives, the RCP was 91.8%. The affinity constant of ^131 I-1H7 with HepG2cells was 1.81 × 10^11L/mol. Conclusion Labeling 1H7 with ^131I by Iodogen method, the product is stable, of a high specific activity, RCP and immune activity.
