摘  要:目的:通过比较联合诱导法和多氯联苯诱导法制备的S9的活化作用,探讨两种S9活化作用的差异对两种试验阳性结果的影响。方法:采用Ames试验方法和染色体畸变试验方法对两种S9活化的阳性结果进行比较。Lowry法测定两种S9的蛋白含量,Omura法测定P450含量。结果:Ames试验和染色体畸变试验中两种S9在阳性组中都显示出明显活化作用,同一条件下,Ames试验中使用两种S9所得阳性结果并无较大差异,染色体畸变试验中两种S9活化的阳性组畸变率间差异无统计学意义(P>0·05)。多氯联苯诱导法S9的蛋白含量13·25g·L-1,P450含量为3·92nmol·mg-1蛋白,联合诱导法S9蛋白含量为15·25g·L-1,蛋白含量P450含量为2·94nmol·mg-1蛋白。结论:两种S9在Ames试验和染色体畸变试验中对相应阳性诱变剂都有明显的活化作用。可以认为两种S9在Ames试验和染色体畸变试验中对于相应的阳性化学物活化作用无较大差异,联合诱导法可以替代多氯联苯诱导法制备肝匀浆S9。Objective: To compare activation of two kinds of S9 which were induced by PCBs and the combination of PB and BF and to investigate effect of the difference of activation of two kinds of S9 on the test result. Methods. The number of revertant colonies of positive contrast group in the presence of different dose of S9 were compared in Ames assay respectively, and mutant frequency of positive control were compared in chromosome aberration, content of protein was determined by the method of Lowry, and content of P450 of two kinds of S9 were determined according to the method of Omura and Sato. Results: In the Ames assay, the number of revertant colonies had dose-response relationship among the dose of S9. When conditions such as strains and the dose of S9 were same, the number of revertant colonies in positive control was at the approximate level in presence of different S9 respectively. Mutant frequency between positive controls in the presence of different S9 had no significant difference in chromosome aberration. Content of protein S9 induced by PCBs was 13.3g·L^-1, content of P450 was 3.92 nmol·mg^-1 pro, content of protein of the other was 15.2 g·L^-1, content of P450 was 2.94 nmol·mg^-1 pro. Conclusion: Two kinds of S9 both show distinct activation in positive control in Ames assay and chromosome aberration. The activation of two kinds of S9 had no significant difference in related strains and positive contrast in Ames as.say and chromosome aberration. The inducing method of combination of PB and BF can take the place of the inducing method of PCBs in the preparation of Liver homogenate S9.
