柔肝冲剂对血瘀型肝纤维化大鼠纤溶酶原激活抑制因子-1影响

摘　　要：目的:探讨柔肝冲剂对血瘀型肝纤维化大鼠纤溶酶原激活抑制因子-1的调控作用。方法:将大鼠随机分为正常组、血瘀组和柔肝组。用酶联免疫吸附法、免疫组化、免疫印迹法等检测血中、肝组织中纤溶酶原激活抑制因子-1(PAI-1)的表达。结果:通过酶联免疫吸附双抗体法、免疫组化、免疫印迹等发现PAI-1在各组大鼠血液中、肝组织中均有表达。PAI-1表达在血瘀组较正常组为高(P<0.01);经柔肝冲剂治疗后PAI-1的表达均有下降(P<0.01)。结论:PAI-1在正常大鼠肝组织中有表达,血瘀型肝纤维化时其在肝组织中蛋白表达增高,柔肝冲剂可降低其表达。柔肝冲剂可能通过影响纤溶酶原激活抑制因子-1起到抗肝纤维化作用。Objective： To study the changes expression of plasminogen activator inhibitor-1 in rats blood and liver tissue with blood stasis type of hepatic fibrosis after treated with liver-softening Granules. Methods： The rats were randomly divided into normal group, blood stasis group and liver-softening Granules group. Detected the level of PAI-1 in serum and the protein expression of PAI-1 in hepatic tissue . Results ： PAI-1 were expressed in serum and liver tissue. The expression of it was higher in blood stasis group than in normal group, and the different were significantly. The expression of it was lower in liver soft Granules group than in blood stasis group, and the different, were significantly. Conclusions： There were expression of PAI-1 in rats＇ serum and liver tissue. The expression of PAI-1 were higher in the type of blood stasis hepatic fibrosis rats. After liver soft-Granules treated, the expression of them were decreased. Maybe liver soft-Granules treated hepatic fibrosis by regulating the plasminogen activator inhibitor-1 and hasten the fibrin dissolve.
