摘 要:目的:研究葡萄籽原花青素提取物(GSPE)治疗乙酸性结肠炎的作用机制。方法:制备大鼠乙酸性结肠炎模型,分别设正常对照组、模型对照组、阳性对照组和GSPE低、中、高剂量组(灌胃);给药8d时分别检测结肠组织中髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)、氧化型谷光甘肽过氧化物酶(GSH-Px)及丙二醛(MDA)的变化。结果:与正常对照组比较,模型对照组MPO含量明显升高(P<0.01);与模型对照组比较,GSPE各剂量组MPO活性、MDA含量明显降低(P<0.05),SOD和GSH-Px活性明显升高(P<0.05)。结论:GSPE治疗乙酸性结肠炎作用机制与其抗炎、抗氧化、提高组织抗氧化能力、减轻脂质过氧化损伤及促进损伤组织的修复有关。OBJECTIVE :To study the mechanism of action of grape seed proanthocynidins extract(GSPE)in treating acetic acid induced colitis gravis in rats. METHODS: Rats model with acetic acid- induced colitis was built, the animals were divided into normal control group, model group, positive control group and GSPE group (which were subdivided into low, medium and high dose subgroups by intragastric administration) . On the 8th day of administration of drugs, the changes of myeloperoxidase(MPO), superoxide dismutase(SOD), glutathione peroxidase (GSH- Px) and malonaldebyde(MDA) in colonic tissue were detected, respectively. RESULTS: The model group had a significantly higher level of MPO content than did the normal control group(P〈0.01) . In GSPE (high, medium, and low dose subgroups) group, MPO activity and MDA content in colon tissue had a significant reduction (P 〈 0.05), but activity of SOD and GSH - Px had a significant increase (P 〈 0.05) as compared with the normal control group. CONCLUSIONS: The mechanism of action of GSPE in treating acetic acid - induced colitis gravis is attributable to its anti - inflammatory action, antioxidation meanwhile enhancing antioxidation of tissues, lessening damage induced by lipid peroxidation and promoting tissue repair.
