不同浓度氢溴酸槟榔碱对结肠平滑肌细胞钙离子移动和收缩力的影响

摘　　要：[目的]观察不同浓度的氢溴酸槟榔碱(Ah)对结肠平滑肌细胞钙离子(Ca2+)移动的影响,以探讨其对平滑肌收缩力的作用。[方法]取培养大鼠结肠平滑肌细胞,以荧光指示剂Fluo-3/AM负载,通过激光共聚焦显微镜(LSCM)动态观察不同浓度Ah对培养的结肠平滑肌细胞Ca2+移动的作用。[结果]Ah在1×10-10、1×10-9、1×10-8、1×10-7、1×10-6、1×10-5、1×10-4mol/L浓度刺激下可使结肠平滑肌细胞[Ca2+]i荧光强度(FI)逐渐升高(P<0.05),而在未加药以及1×10-13、1×10-12、1×10-11mol/L浓度刺激下对[Ca2+]iFI无影响(P>0.05);在1×10-3mol/L浓度刺激下细胞膜迅速发生破裂;应用阿托品预处理,则FI从4.28±1.51降至3.83±1.40(P>0.05)。[结论]Ah可使结肠平滑肌细胞内[Ca2+]i逐渐升高,阿托品可阻断Ah对平滑肌的收缩作用。[Objective] To study the effects of arecolinehydrobromide （Ah） at different concentration on calcium mobilization in cultured smooth muscle ceils of rat clone. [Methods] The fluorescent intensity （FI） of intercellular calcium concentration （[Ca^2＋ ]i） was measured with confocal microscope in cultured smooth muscle cells of rat clone loaded with Fluo-3/AM. [Results] At the concentration of 11×10^-10、1×10^-9、1×10^-8、1×10^-7、1×10^-6、1×10^-5 and 1×10^-4mol/L, FI of [Ca^2＋]i in smooth muscle cells elevated gradually by Ah （ P〈0. 05 ） respectively. However, at the concentration of 1×10^-13、1×10^-12、1×10^-11 mol/L, Ah had no effect on the the FI of [Ca^2＋ ]i in smooth muscle cells. Cell membrane ruptured rapidly at the concentration of 1 × 10^-3 mol/L of Ah. When pretreated by atropine,Ah （1 × 10^-4 mol/L） did not increase FI significantly. [Conclusion]Ah can increase [Ca^2＋]i in smooth muscle cells gradually. Atropine has the antagonist effect on Ah.
