无孔单分散亲水性树脂的合成及其在生物大分子分离中的应用

摘  要:建立了一条从分散聚合法制备种子到一步种子溶胀聚合法制备粒径5.0μm的无孔单分散亲水性交联聚甲基丙烯酸环氧丙酯树脂的新方法,对其表面用新的化学方法改性后得到一种新型的弱阳离子交换色谱填料.实验考察了流动相的流速及有机溶剂等对蛋白质保留的影响.结果表明,该填料具有良好的色谱性能,在流速为5 mL/min时4种蛋白可在3 min内达到基线分离,将其应用于鸡蛋清中溶菌酶的快速分离纯化,取得了较好的效果.5 μm non-porous monodisperse poly(glycidylmethacrylate co ethylenedimethacrylate) beads were synthesized by one-step swelling and polymerization method. By using this media, a weak cation exchange (WCX) stationary phase for HPLC was synthesized by a new chemical modification method. Various factors influencing protein separation, such as flow-rate and organic solvent in the mobile phase were studied. The resins as matrix materials possesss excellent properties of separation proteins. Four proteins were completely separated in 3.0 rain using the synthesized WCX stationary phase. The WCX resin was used for the rapid separation and purification of lysozyme from egg white with only one step. The purifity of the purified lysozyme was higher than 92.0%.
