摘 要:目的 建立HPLC法测定血浆中氨茶碱浓度的方法。方法 采用反相高效液相色谱法测氨茶碱血药浓度。C18柱,流动相为甲醇-水(32:68,v/v),检测波长275nm。结果 在5~25mg·L^-1浓度范围内线性良好,r=0.9997最低检测限为1mg·L^-1,平均回收率为94.12%~108.59%,日内误差与日问误差均小于8%。结论 该法操作简便、快速、准确,可用于氢茶碱的药物监测。Aim To develop RP-HPLC method for the determination of aminophylline in serum. Methods Chromatographic separation has been achieved on C18 column with methanol:water (32: 68,V/V) as the mobile phase. Detection was set at UV 275nm. Result The calibration curve was linear between 5 mg ·L^-1 and 25 mg ·L^-1 with a regression coefficient of 0. 9997. The lower limit of detection of aminophylline in serum was 1 mg ·L^-1. The recovery rate was 94.12% ~ 108.59%. Tnter-day RSD and intra-day RSD of aminophylline were less than 8%. Conclusion The method was simple,rapid and accurate. It could be successfully used for TDM.
