摘  要:目的 研究亚硒酸钠、氯化镉及其联合作用对大鼠肝脏端粒酶逆转录酶(TERT)mRNA表达的影响。方法 健康雄性SD大鼠随机分组,每组动物5只。对照组,硒组(2.5、5和10μmol/kg Na2SeO2)、镉组(5、10和20μmol/kgCdCl2)以及硒镉联合作用组。染毒48h后取出肝脏,用RT-PCR方法检测TERT基因的表达。结果 与对照组比较,3个剂量硒组的TERT mRNA虽有增高,但P〉0.05,而3个剂量的镉组TERT mRNA表达均增多(P〈0.01);在硒镉联合作用组中,虽然3个剂量的硒均可分别使3个剂量的镉诱导的大鼠肝脏TERT mRNA表达下降,但仅在2.5μmol/kg Na2SeO3+5μmol/kgCdCl2组、2.5μmol/kg Na2SeO3+10μmol/kg CdCl2组和5μmol/kg Na2SeO3+10μmol/kg CdCl2组,与相应剂量的镉组比较,TERT mRNA水平降低,差异具有显著性。10μmol/kg Na2SeO3+5、10和20μmol/kg CdCl2联合作用组与对照组相比,TERT mRNA水平均升高,与对应镉组相比,差异无显著性。结论 镉在5~20μmol/kg的剂量条件下可以诱导大鼠肝脏高表达TERT mRNA,而一定剂量的硒对一定剂量镉引起的TERT mRNA的表达具有一定的拮抗作用。Objective To study the effects of sodium selenite( Na2 SeO3 ), cadmium chloride( CdCl2 ) and their joint action on expression of telomerase reverse transcriptase(TERT) mRNA in rat liver. Methods Male SD rats were divided randomly to 16 groups, each group had 5 animals. The groups include : control group, Se groups ( Na2 SeO at the doses of 2.5,5 and 10 μmol/kg, Cd groups (CdCI: at the doses of 5,10 and 20 μmol/kg, Se( Na2 SeO3 at the doses of 2.5,5 and 10 μmol/kg + Cd( CdCl2 at the doses of 5,10 and 20 μmol/kg groups. After 48 h of the first injection ,the expression of TERT mRNA was measured with Reverse transcription polymerase chain reaction (RT-PCR). Results Although both Se at the doses of 2.5,5 and 10 μmol/kg and Cd at the doses of 5,10 and 20 μmol/kg could increase rTERT expression in rat liver,compared with control group, statistical analysis yielded P 〈 0.01 only in Cd groups. In the joint action groups of Se and Cd, Se at the doses of 2.5,5 and 10 μmol/kg could decrease rTERT over expressions induced by Cd at the doses of 5,10 and 20 μmol/kg respectively, but statistical analysis showed that rTERT expressions in 2.5 μmol/kg Na2 SeO3 + 5 μmol/ kg CdCl2 group,2.5 μmol/kgNa2SeO3 + 10 μmol/kgCdCl2 group and 5 μmol/kgNa2SeO3 + 10 μmol/kgCdCl2 group were lower than those of correspondent Cd group respectively and significantly. The expression of rTERT mRNA in 10 μmol/kgNa2 SeO3 + 5,10 and 20 μmol/kgCdCl2 groups were no significant different from correspondent Cd groups, but higher than that of control significantly. Conclusion It was suggested that cadmium at the doses of 5 μmol/kg~ 90 μmol/kgcan induce overexpression of TERT mRNA in rat liver. Selenium at the doses of 2.5 ~ 5 μmol/kg have antagonistic effect on overexpression of TERT induced by Cd in rat liver.
