摘  要:目的研究镉(Cd)致大鼠血中几种免疫介质、脾脏淋巴细胞转化和NK细胞活性变化,探讨慢性Cd接触对大鼠免疫介质释放和免疫细胞活性的影响。方法选择健康SD雌性成年大鼠54只,随机分成对照组,低剂量氯化镉组,高剂量氯化镉组。对照组每天给予普通全价饲料,低、高Cd组分别饲喂含镉2和10 mg/kg的全价饲料,连续9周。分别在第3、6和9周采集血样和脾脏进行分析。结果在整个实验期内,Cd诱导血浆环磷酸腺苷(cAMP)水平升高,且第3周高Cd组及第9周低Cd组与对照组差异显著;Cd接触大鼠血浆NO均升高,且高Cd组显著高于对照组(P<0.05);Cd诱导血浆白细胞介素-2(IL-2)水平下降、肿瘤坏死因子-α(TNF-α)水平上升,但与对照组差异不明显。慢性Cd接触大鼠脾脏淋巴细胞转化刺激指数均显著低于对照组,NK细胞活性随剂量增加和时间的延长而明显下降。结论慢性Cd接触诱导大鼠免疫细胞活性抑制,一些免疫介质发生紊乱和较大变化可能在介导免疫细胞功能活动低下发生过程中有重要作用。Objective Changes of some blood immune-mediators as well as spleen lymphocyte transformation and natural killer (NK)cell activities in cadmium(Cd) induced rats were studied to discuss the effects of chronic Cd exposure on immune-mediator releases and immunocyte activities in rats. Methods Fifty-four healthy SD female rats were chosen and divided randomly into control group, low dose cadmium chloride( CdCl2 ) group ( low-Cd group) and high dose CdCl2 group ( high-Cd group). During 9 weeks experimental period, the common complete diet was orally provided dally in control group, and the rats in low-Cd and high-Cd groups were given the diets with 2 and 10 mg of Cd per kg feed, respectively. The specimens of blood and spleen were collected for analysis at the 3rd, 6th and 9th week. Results During the entire experimental period, the Cd-induced elevation of plasma cAMP levels was observed, which was significantly higher in high-Cd group at the 3rd week and in low-Cd group at the 9th week than in control group; Plasma nitrous oxide (NO) of Cd-exposed rats increased and high-Cd group was markedly higher than control group( P 〈 0.05) ; Plasma interleukin-2(IL-2) decrease and tumor necrosis factor-α(TNF-α) increase of Cd-exposed rats had no significant difference compared to those of control animals, the stimulation index of spleen lymphocyte transformation by Cd was significantly lower than that of control group; Spleen NK cell activity significantly dropped with dosage increase and time elongation of Cd exposure. Conclusion chronic Cd exposure induces the inhibition of immunocyte activities in rats. The derangement and violent changes of some immune-mediators may play an important role in mediating the underactivity of immunocyte function.
