摘 要:目的通过观察电针不同穴组对急性脑缺血大鼠72h后脑组织神经肽Y(neuroptide Y NPY)含量和NPYm RNA表达的影响,比较电针不同穴组的针刺效应并探讨其抗脑缺血损伤的作用机制。方法将雄性SD大鼠25只随机分为假手术组,模型组,人中、百会组,肝俞、肾俞组,曲池、足三里组,每组5只;右颈外动脉插入线栓法建立大鼠大脑中动脉局灶性脑缺血(MCAO)模型。采用放射免疫法测NPY,RT-PCR半定量分析法测NPYm RNA的表达。结果大鼠MCAO急性脑缺血72h后,电针人中、百会组及肝俞、肾俞组使脑组织NPY含量及NPYm RNA的表达明显低于模型组(P<0.05);两组之间比较,差异无统计学意义(P>0.05);电针曲池、足三里组对上述指标均无明显影响(P>0.05)。结论电针人中、百会组及肝俞、肾俞组可抑制脑组织N PYm RNA的表达,降低N PY的含量,对抗脑缺血损伤,且两穴组针刺效应相近而优于曲池、足三里组。Objective To compare the acupuncturing effects between the defferent point groups and explore the mechanism of electro-acupuncture for acute cerebral ischemia injury via obeservation of effect on NPY and its gene expression in the brain tissues of rats with acute cerebral ischemia after 72 hours by electro-acupuncturing different points. Methods 25 male SD rats were randomly divided into 5 groups, i.e. pseudo-operation, model, points Renzhong and Baihui, points Ganshu and Shenshu and Quchi and Zusanli group, to establish the MCAO model. NPY was detected with radioimmunoassay; NPYmRNA expression was measured with RT-PCR. Results After establishment of MCAO model for 72 hours, electro-acupuncturing Renzhong and Baihui group and Ganshu and Shenshu group, the content of NPY and it's expression of NPYmRNA in the brain tissues were lower than that of model group(P〈0.05); com- pared the two groups, the dfference was no meaningful (P〉0.05); In the group of electroacupuncturing Quchi and Zusanli, there was no remarkable influence (P〉0.05). Conclusions Electro-acupuncturing in the group Renzhong and Baihui and group Ganshu and shenshu can inhabit expression of NPY mRNA, reduce the content of NPY and resist against cerebral ischemia lesion and the effects are similar in these two groups and better than that in group Ouchi and Zusanli.
