摘 要:目的探讨ICAM-1在大鼠弥漫性脑损伤后脑水肿中的作用。方法采用Marmarou等人的方法建立大鼠弥漫性脑损伤模型,S-P免疫组化法测定ICAM-1蛋白在外伤后不同时间脑组织中的表达,干湿重法测脑组织含水量,组织切片HE染色观测炎性细胞浸润情况。结果伤后6h,脑组织中ICAM-1蛋白表达明显升高,72h达到高峰,其后下降,7d时仍明显高于假手术对照组(P<0.05)。伤后6h、12h、24h、48h、72h和7d时,脑组织含水量亦明显高于假手术组(P<0.05),同时伤后脑组织中炎性细胞大量聚集。结论大鼠脑外伤诱导了ICAM-1蛋白的表达,ICAM-1通过介导炎性细胞的浸润可能参与了伤后脑水肿的形成过程。Objective To explore the role of intercellular adhesion molecule-1 (ICAM-1) in brain edema after diffuse brain injury in rats. Methods The model of diffuse brain injury in rats was established by Marmarou et al technique. The expression of ICAM-I protein at different time after the injury was determined by immunohistoehemistry. The water content in the brain tissues was determined. The pathological examination of the injuried brain tissues was performed. Results The expression of ICAM-1 protein was signifi-cantly increased at the 6th hour after the injury (P〈0.05), peaked at the 72nd hour and then was decreased. The expression of ICAM-1 in the traumatic group still was significantly higher than that in the control group on the 7th day after the injury (P〈0.05) The water content in the brain tissues at the 6th, 24h, 24th, 48th and 72nd hours, and on 7th day after brain injury was significantly higher than that in the control group (P〈0.05). At the same time, a great deal of inflammatory cells gathered in the brain tissues. Conclusions Ditthse brain injury can induce the expression of ICAM-1 in the posttraumatic brain tissues. It is suggested that ICAM-1 may be involved in the development of brain edema by mediating the infiltration of inflammatory cells into the posttraumatie brain tissue.
