摘 要:目的研究体外循环围手术期不同时点Toll样受体4(TLR4)的表达变化。方法选取20例体外循环下心内直视手术病人,分别于麻醉诱导前(T1)、体外循环前(T2)、体外循环后即刻(T3)、体外循环后4 h(T4)、体外循环后24 h(T5)抽取外周静脉血,分离单核细胞,用逆转录聚合酶联反应(RT-PCR)法检测TLR4的表达变化。结果与术前比较,体外循环前、体外循环结束后即刻单核细胞TLR4的表达下调,体外循环结束后4 h表达上调,体外循环结束后24 h进一步升高。结论体外循环围手术期,TLR4的表达呈现动态变化,TLR4可能与体外循环术后炎症反应等病理生理有一定关系。Objective to study the Expression changes of TLR4 in peripheral blood monocyte during cardiopulmonary bypass(CPB). Methods 20 patients with heart diseases underwent CPB were selected for this study. The expression of TLR4 were measured by RT-PCR. Blood samples were taken through vein at 5 different time points-preoperative, before CBP, 0 hours, 4hours and 24 hours after CPB. Results The expression of TLR4 was down-regulated at T2 and T3,and upregulated at T4 and T5 ,and peaked at T5. Conclusion the Expression of TLR4 during cardiopulmonary bypass at 5 different time points is discrepant clearly,may be TLR4 is related to the inflammatory response after CPB.
