摘  要:目的研究红毛五加多糖提取及含量测定的最佳条件。方法用正交实验法分别优选苯酚硫酸比色法测定红毛五加多糖含量的最佳条件和红毛五加多糖的最佳提取工艺。结果最佳测定条件为;苯酚用量为0.5mL,浓硫酸用量为6mL,反应时间为60min;最佳条件下,在22-90μg·mL^-1多糖含量与吸收度有良好的线性关系。测得红毛五加中多糖含量为8.8%。在此基础上,用正交实验法优选红毛五加多糖的提取条件,提取工艺的最佳条件为:煎煮0.5h、煎煮3次,加水量10mL·g^-1,乙醇浓度70%。结论本研究为红毛五加多糖的提取和含量测定的条件提供了参考,为红毛五加的进一步研究提供了基础。OBJECTIVE To investigate the optimum conditions for the determination and extraction of Acanthopanax giraldii harms polysaccharides. METHODS Orthogonal design was used to optimize the conditions for the determination and extraction of Acanthopanax giraldii harms polysaccharides (AGP) by phenylhydrate-sulfurie acid colorimetry. RESULTS The best conditions were as follows: phenylhydrate, 0. 5 mL; sulfuric acid, 6 mL; and reaction time, 60 min. Within the range of 22 mg ·L ^-1 to 90 mg ·L ^-1, there was a good linear relationship between the con centration of AGP and the absorbance . The AGP in Acanthopanax giraldii harms was 8.8%. Based on the results, the condition for extracting of AGP were optimized by orthogonal design as follows: boiling for 0. 5 h for 3 times, the volume of water for extracting was 10 mL per gram of Acanthopanax giraldii harms and the precipitating concentration of methanol was 70%. CONCLUSIONS The study provides references for the determination and extraction of Acanthopanax giraldii harms polysaecharides and groundwork for its further investigation.
