摘  要:表达序列标签是一种快捷、高效的揭示基因组信息的方法。本研究以蓝塘猪与大白猪差异表达的5条EST在各组织中的表达情况为目的,采用大白猪、蓝塘猪为实验材料,从成年种猪的10个组织中提取总RNA,运用RT-PCR方法分析,结果显示5条EST在猪的多数组织中均有表达;同源性比对后发现,在5条差异显示cDNA中,EST1、EST2与已知序列无同源性;电子表达谱分析表明EST1、EST2、EST4及EST5在骨骼肌、肝脏组织中有表达。这为进一步研究该EST的功能奠定基础。The generation of expressed sequence tags(ESTs) has been proved to be a rapid and efficient approach to disclose the information of genome. In this paper, in order to get a better understanding of 5 pig EST expression situation with Lantang and Yorkshire of all organs, the totle RNA extracted from 10 organs, analyse the expression of the ESTs among the different tissues by RT-PCR method, the result is: 5 ESTs are expressed in the majority of the pig organs; Homologous analysis of these sequence with those available in GenBank indicated that EST1 and EST2 are novel ESTs. EST3 was identical to Homosapiens caveolin 1, EST4 was identical to mouse methionine aminopeptidase-likel gene, EST5 was identical to Sus Serofa insulin-like-growth factor 2 preproteine genes. The electron expresses pattern analyses that indicates EST1 , EST2 , EST4 and EST5 are expressed in the muscles, liver. The research established the foundation for function of studying this EST further.
