摘 要:目的探索制备适用于超声治疗的阴离子脂膜超声微泡,评价其理化性质。方法通过改变配方中脂质成分比例以及聚乙二醇(PEG)的用量,制备出三种配方的脂膜超声微泡,分别检测微泡大小形态、浓度、表面电位、PH值、粒径及分布。结果三种微泡的浓度约(1-2)×10^10/ml;粒径均值分别为1066.6nm、1552.5nm、1605.4nm;pH值分别为:蒸馏水中2.94,2.63,2.52,生理盐水中3.77,2.60,2.55;微泡表面电位均值分别为:蒸馏水中-33.8mV、-57.6mV、-66.7mV;生理盐水中0.1mV、-37.5mV、-37.8mV。结论通过增加脂质成分中负电荷磷脂的比例,可以制备出表面带有更多负电荷的阴离子超声微泡,为超声介入治疗提供重要的工作平台。Objective The purpose of this study was to prepare anionic lipid- coated microbubbles with a net negative charge and to assess its physiochemical properties. Methods Two configurations of DSPC and DPPA with different proportions were designed to prepare the negative charged microbubbles, with or without PEG component. The bubbles concentrations were counted under light microscopy and the pH value, the surface potential and the microbubbles size were assessed by the Zeta SIZER 3000HSA. Results The bubbles concentrations,stability in vitro of the three microbubbles nearly have no difference,but their electronic properties have obvious difference. All the concentrations were about (1 - 2) × 10^10/ml. The bubbles average diameters were 1 066.6 nm, 1 552.5 nm, 1 605.4 nm, respectively. The pH values were 3.77, 2.60, 2.55 in saline and 2.94, 2.63 and 2.52 in distilled water. The electronic potentials Were 0. 1mV, - 37.5 mV, - 37.8 mV in saline and - 33.8 mV, - 57.6 mV, - 66.7 mV in distilled water. Conclusion Anionic lipid microbubbles with a net negtive charge could be prepared by changing the pmportian of lipids. This design of a new anionic microbubbles was specific for therapeutic purpose.
