RCS大鼠视网膜色素变性过程中内源性干细胞激活的初步研究

摘　　要：目的研究遗传性视网膜色素变性大鼠(Royal College of Surgeon’s rat,RCS rat)在视网膜变性发展过程中是否有内源性视网膜干细胞(retinal stem cells,RSCs)激活。方法RCS-p+(视网膜含色素的变性大鼠)按出生后视网膜变性的发展状况分为变性早期(RCS15d)、中期(RCS30d)、晚期(RCS90d)3个时相点,分别采用相应时期的RCS-rdy+p+(视网膜含色素的正常大鼠)作为对照组(control groups,C15d,C30d,C90d),每组4只8眼,对各组视网膜切片进行免疫荧光组织化学(抗Chx10)实验,以鉴定视网膜干细胞;进一步采用Western blot检测各组视网膜神经上皮总蛋白中Chx10表达。结果①各正常组及变性15d大鼠视网膜各层抗Chx10免疫荧光检测均为阴性,变性中、晚期视网膜光感受细胞层部位Chx10免疫荧光阳性;②Western blot检测各变性组视网膜神经上皮总蛋白中Chx10均有表达,但变性中期Chx10表达显著高于早期和晚期(P<0.05)。结论RCS大鼠变性过程中视网膜感光细胞层出现视网膜干细胞激活。Objective To investigate whether endogenous retinal stem cells can be activated in the retina of Royal College of Surgeons （RCS） rats during the onset and development of retinitis pigmentosa. Methods The RCS-p ＋ rats with inherited retinal dystrophy were divided into 3 groups： the initial stage group （ 15-day RCS rats）, the mid-stage group （30-day RCS rats） and the advanced stage group （90-day RCS rats） according to the severity of degeneration （n --4 in each group）. RCS-rdy ＋ p ＋ rats without retinal degeneration served as controls, and divide into three groups （15-day control, 30-day control, 90-day control） matched with RCS-p ＋ rats. A transcription factor （Chxl0） expressed by embryonic retinal progenitors was detected using immunofluorescence and Western blotting. Results All of the retinal layers in the three control groups and in the 15-day RCS rats did not express Chx10, while the positive expression was observed in the photoreceptor layers of the 30-day and 90-day RCS rats. Chxl0 protein could be detected by Western blotting in all RCS groups, but expressed higher in 30-day RCS rats than in 15-day and 90-day RCS rats （P 〈 0.05）. Conclusion Endogenous retinal stem ceils can be activated in the photoreceptor layer of RCS rat retina during the onset and development of retinitis pigmentosa.
