摘  要:目的:检测肌细胞增强因子2(MEF-2)在兔后肢侧支血管形成过程中的表达。方法:结扎兔左侧股动脉,术后1周和4周分别处死动物,冰冻切片进行MEF-2和Ki67(细胞增殖标志物)免疫荧光染色,共聚焦显微镜观察。结果:正常血管MEF-2仅在中膜有微弱表达;在生长发育的侧支血管壁,MEF-2全层的表达均显著增强;而在发育成熟的侧支血管MEF-2的表达接近于正常血管水平。Ki67和MEF-2的双重免疫组化显示二者在细胞中的表达有重叠。结论:在兔后肢动脉生成过程中MEF-2表达上调,可能通过参与细胞的增殖及MCP-1的调节促进侧支血管生成。Objective: To investigate the expression of MEF-2 during collateral vessel arteriogenesis in rabbit hind limb. Methods: Left femoral artery was ligated in rabbit with one knot. Then the growing collateral vessels were obtained at week 1 and 4 postsurgery; cryosections were prepared and immunohistochemistry was done. The sections were viewed with a confocal microscope. Results: Only weak expression of MEF-2A was detected in the SMCs of the tunica media in normal arterial vessels. Expression of MEF-2A was significantly enhanced in all layers of the wall in growing collateral vessels. However, the expression of MEF-2A in mature collateral vessels restored to the normal level. Dual immunostaining of MEF-2A with Ki67 confirmed the colocalization of these 2 proteins in proliferating cells. Conclusion: MEF-2 expression is upregulated in the growing collateral vessels of rabbit hind limb, which may contribute to collateral vessel growth via mediating cell proliferation and MCP-1 expression.
