摘 要:目的探讨孕期膳食补锌对胎盘及母鼠小肠中IGF-II mRNA表达的影响。方法18只SD孕鼠随机分为正常对照组(ZC)和补锌组(ZS)。两组进食常规饲料,ZC组(n=9)饮用去离子水,ZS组(n=9)饮用补锌水(含锌1.26 mmol/L)。分别于孕9.5 d和孕17.5 d时取胎盘和小肠。以半定量聚合酶链反应(RT-PCR)测定胎盘和母鼠小肠中IGF-II mRNA的相对表达量。结果ZC组孕17.5 d时,胎盘IGF-II mRNA的相对表达量明显高于孕9.5 d(P<0.05),呈现出发育性增高。孕9.5 d和孕17.5 d时,ZS组胎盘及小肠IGF-II mRNA相对表达量均显著高于ZC组(P<0.05)。结论妊娠不同时期膳食锌摄入水平增高显著上向调控胎盘和母体小肠中IGF-II mRNA的表达。Objective To investigate the effect of zinc supplementation on the expression of IGF-Ⅱ mRNA in placenta and small intestine of rats during pregnancy. Methods 18 healthy pregnant SD rats were randomly divided into normal control (ZC) and zinc supplement group (ZS). Rats in both groups were fed with the same normal commercial diet. Rats in ZC group drank deionized distilled water and those in ZS group drank the water supplemented with zinc of 1.26 mmol/ L. Placenta and small intestine were taken at gestational day 9.5 and 17.5, respectively. The expression of IGF-Ⅱ mRNA was examined by RT-PCR. Results On gestational d17.5, the expression of placental and small intestinal IGF-Ⅱ mRNA in ZC group was significantly higher than that on gestational d9.5. In ZS group on the gestational d9.5 and d17. 5, the expression of IGF-Ⅱ mRNA in placental and small intestine was significantly higher than that in ZC group. Conclu- sion Dietary zinc supplementation during pregnancy significantly up-regulated the expression of IGF-Ⅱ mRNA in placenta and small intestine of pregnant rats.
