摘 要:目的探讨主动脉-性腺-中肾(AGM)区来源的基质细胞诱导胚胎干细胞(ESC)向成血-血管干细胞分化的促进作用。方法从孕11 d的小鼠胚胎AGM区分离、培养基质细胞,鉴定后用作支持细胞,与小鼠胚胎干细胞共同培养,通过原始细胞集落(BL-CFC)的形成情况及流式细胞仪检测CD34+/Flt-1+细胞比例,评价AGM区基质细胞对小鼠ESC-D3细胞系向成血-血管干细胞的定向诱导作用。结果在AGM区基质细胞的支持下,由ESC来源的CD34+细胞由(12.71±1.76)%增加到(39.71±3.76)%,并且CD34+/Flt-1+细胞比例明显提高,达到(26.59±1.77)%;同时由ESC来源的代表成血-血管干细胞的BL-CFC集落增加了3倍。结论AGM区基质细胞可促进胚胎干细胞向成血-血管干细胞分化,探明其机制对研究造血及血管发育机制有积极意义。Objective To explore the role of aorta-gonad-mesonephros (AGM)-derived stromal cells in promoting the differentiation of embryonic stem cells (ESCs) into hemangioblasts. Methods The murine stromal cells were separated and cultured from the AGM region of a 11 daypostcoitum (dpc) mouse embryo. After phenotypic analysis, the stromal cells were cocultured with the ESCs. ESC-derived and cytokine-induced hemangioblasts were detected by flow cytometry and colony cells as say. Results Supported by AGM-derived stromal cells, the ratio of ESCs-derived CD34^+ was increased from (12.71±1.76) % to (39.71±3.76) %, and CD34^+/Flt-1^+ cells were markedly increased to (26.59± 1.77) %. The number of BL-CFCs produced by the hematopoietic precursor cells induced by co-cuhure with AGM stromal cells was increased by 3 times as compared with the control cells. Conclusion AGM-derived stromal cells, which could promote the differentiation of ESCs into hemangioblasts, play an active role both in hematopolesis and angiogenesis.
