姜黄素和干扰素γ抑制慢性髓系白血病STAT5基因表达的研究

摘  要:目的探讨姜黄素和干扰素γ(IFN-γ)对慢性髓系白血病(CML)K562细胞STAT5基因表达的抑制作用及其作用机制。方法运用噻唑蓝(MTT)比色法检测细胞增殖活性。运用逆转录聚合酶链式反应(RT-PCR)检测细胞STAT5 mRNA的变化。运用激光共聚焦显微镜和免疫印迹法检测STAT5蛋白的表达。结果①姜黄素或IFN-γ作用K562细胞24 h,K562细胞增殖活性明显受到抑制,STAT5 mRNA和STAT5蛋白表达降低。②姜黄素与IFN-γ联合干预K562细胞24 h,细胞增殖活性、STAT5 mRNA和蛋白的表达均显著低于姜黄素组(均P<0.01)。结论姜黄素和IFN-γ联用能明显抑制慢性髓系白血病细胞增殖以及STAT5基因表达。Objective To investigate the inhibitory effect of cureumin and IFN-γ on the expression of STAT5 gene in chronic myeloid leukemia (CML) cell line (K562 cell line) and the mechanism. Methods Cell proliferation was studied by tetrazolium dye assay. The expression of STATS mRNA was detected by RT-PCR and that of STATS protein by immunoblotting and confocal laser scanning microscopy. Results When K562 cells were treated with cureumin or IFN-γ for 24 h, the expression level of STATS mRNA and protein was down-regulated obviously. The expression level of STAT5 mRNA and protein was decreased in the group of combined use of eureunin with IFN-γ, statistically less than that in eurcumin group (P〈0.01). Conclusion The combined use of curcunin with IFN-γ inhibits the cellular proliferation and decreases the expression of STAT5 gene in CML cells.
