葛根素对损伤内皮细胞白介素-8释放的影响

摘　　要：[目的]研究葛根素对动脉粥样硬化发病过程中血管内皮细胞分泌细胞因子的影响,探讨其抗动脉粥样硬化的机制。[方法]通过建立同型半胱氨酸(HCY)诱导的细胞损伤模型,运用放射免疫方法,检测不同浓度葛根素处理的人脐静脉内皮细胞系-304(ECV304)细胞培养液中白介素-8(IL-8)含量。[结果]HCY明显增加培养上清中趋化因子IL-8的含量;葛根素高剂量组则可减少因HCY诱导损伤内皮细胞趋化因子IL-8的分泌。[结论]葛根素高剂量组可通过抑制损伤的ECV304细胞分泌IL-8,从而保护损伤的血管内皮细胞,发挥抗动脉粥样硬化作用。[Objective] To study the influence of Puerarin on the secretion of cytokine from the vascular endothelial cell during the course of atheroosclerosis and explore its mechanism of anti-atherosclerosis. [Methods] The contents of IL-8 in the cell culture fluid treated by various concentration were detected in the cell model induced by homocystein （HCY） and radio-immunity method. [Results] HCY could increase the contents of IL-8 in the fluid apparently, high dose of Puerafin ould reduce the secretion of IL-8. [Conclusions] High dose Puerarin can reduce the secretion of IL-8 by inhabiting injured ECV304. Thereby it can protect the injured endothelial cell and has effect on anti-atherosclerosis.
