摘  要:采用同工酶技术和RAPD技术对宁波黑斑口虾蛄的遗传多样性进行了分析,采用聚丙烯酰胺凝胶电泳方法(PAGE)对LDH、MDH、MEP、EST、ADH、GDH、GLDH、AMY、POD、SDH等10种同工酶进行了群体生化遗传分析,共检测了29个基因座,其中11个基因座表现为多态,其多态位点比例为37.93%,平均杂合度为0.1470;采用RAPD技术对其遗传多样性进行检测,用24个随机引物共检测出129个位点,其中多态位点为102个,占79.06%,个体间遗传相似系数为0.3106,遗传变异度为0.6894,表明黑斑口虾蛄的遗传多样性状况良好。Randomly amplified polymorphic DNA (RAPD) analysis and isozyme technique were used to examine genetic variation in 34 mantis shrimp Oratosquilla kempi samples collected in Ningbo, Zhejiang province by 24 selected arbitrary primers and 10 isozymes. There were 129 polymorphic RAPD fragments ranging in size from 200 to 2000 bp in the mantis shrimp with 102 polymorphic loci ( accounting for 79.06% in total polymorphic amplified fragments). The basic electrophoretic patterns of the isozymes showed that 29 gene loci were scored, in which 11 gene loci including Ldh-1 ( alleles, 100,120), Adh-1 ( alleles, 100,105 ), Sdh-1 ( alleles, 100,110), Sdh-2 ( al- leles, 100, 110), Est-2 (alleles, 100, 105), Mdh-1 (alleles, 80,100), Mep-3 (alleles, 95,100,110), Mep-4 ( alleles, 100,105,110, 115 ) Gldh ( alleles, 90, 100), Gdh ( alleles, 100,110) and Amy-4 ( alleles, 100,105 ) were polymorphic, accounting for 37.93%. The value 0. 1470 was found for average heterozygosity, indicating that the natural mantis shrimp resources were in good condition with higher gengtic variability, and effective measure should be taken to maintain the good status.
