摘 要:以两个苜蓿品种新牧1号和新疆大叶苜蓿无菌苗的幼叶为外植体,研究了4种诱导培养基对愈伤组织的影响,增殖培养基中添加水解酪蛋白对愈伤组织增殖的影响,以及分化培养基中KT对愈伤组织分化的影响。结果表明:①叶片愈伤组织诱导培养基以MS+2,4-D 2.00 mg/L+NAA 1.00 mg/L+KT 1.00 mg/L为宜,新牧1号和新疆大叶苜蓿的出愈率分别为100%,95.5%;②增殖培养基中附加CH 0.15 g/L有利于新牧1号愈伤组织的生长,附加CH 1.00 g/L有利于新疆大叶苜蓿的愈伤组织的生长;③分化培养培养基中附加KT 4.00 mg/L,两品种的分化率最高分别为新牧1号25.0%、新疆大叶苜蓿13.2%。The leaves of the two varieties of Medicago sativa L. cv. Xinjiang Daye and Medicago varia Martyn. cv. Xinmu No. 1 were cultured in four kinds of induction medium to study the induction of callus, effects of CH on enrichment, and differentiation callus in differentiation media with KT. The results were as follows.①MS+2,4 D 2.00 mg/L+NAA 1.00 mg/L+KT 1.00 mg/L was the better induction medium,the callus ratio of Medicago varia Martyn. cv. Xinmu No. 1 and Medicago sativa L. cv. Xinjiang Daye separately were 100%,95.5% respectively; ②Adding CH 0.15 g/L in enrichment medium was bene fit to the growth of Medicago varia Martyn. cv. Xinmu No. 1, CH 1.00 g/L was propitious to the growth of Medicago sativa L. cv. Xinjiang Daye;③With KT 4.00 mg/L in differentiation medium, the differentiation rate of Medicagovaria Martyn. cv. Xinmu No. 1 was 25.0% and Medicago sativa L. cv. Xinjiang Daye was 13.2%.
