摘 要:目的:建立一种HPLC/UV/MS/MS清开灵注射液(板蓝根,栀子,金银花等)指纹图谱研究方法,为其质量控制提供新思路。方法:采用Phenom enex Luna C18色谱柱(250 mm×4.6 mm,5μm),流动相分别为0.1%甲酸水溶液和乙腈梯度洗脱,流速0.5 mL/m in;采用电喷雾离子阱质谱(ESI-ITMS)分析,正、负离子二级扫描。结果:得到分离度和重现性良好的HPLC/UV/MS/MS指纹图谱,对其指纹图上13个色谱峰进行了初步定性,成功鉴别了新绿原酸、绿原酸和异绿原酸3个异构体。结论:本方法建立了清开灵注射液HPLC/UV/MS/MS指纹图谱,为中药复杂体系的分析及其质量控制提供了一种有效、可靠的模式。AIM : To develop a method of the HPLC/UV/MS/MS fingerprints of Qingkailing Injection ( Radix Isatidis, Fruetus Gardeniae, Flos Lonieerae Japonieae, etc. ) and provide a novel conception for its quality control. METHODS: Separation was performed on Phenomenex Luna C ls analytical column (250 mm × 4.6 mm, 5μm). The mobile phases consisted of water containing 0.1% formic acid and aeetonitrile was used as gradient elute. The flow rate was 1 mL/min. An Electrospray Ion Trap mass spectrometer was utilized for qualitative analysis and both positive and negative secondary ion scan mode were applied. RESULTS: Perfect fingerprints with excellent separation and reproducibility were obtained and 13 compounds were identified. Especially three isomers, including Neochlorogenie acid, Chlorogenie acid and Isochlorogenie acid, were differentiated successfully. CONCLUSION: The method is suitable for establishing the HPLC/MS/MS fingerprints of Qingkailing Injection and hence could be used as an effective and credible pattern for analyzing traditional Chinese medicine as well as its quality control. Moreover, based on this approch, further traditional Chinese medicine research can be performed.
