摘 要:目的:观察人参二醇皂甙对内毒素诱导休克大鼠模型肺组织的保护作用。方法:实验于2003-05/12在吉林大学基础医学院机能科学实验中心实验室完成。①选用健康Wistar大鼠100只,雌雄不拘,鼠龄2.5~3月。按随机抽签法将大鼠分为5组:模型组,地塞米松组,人参二醇皂甙低、中、高剂量组。每组再分别于休克后120和240min处死10只大鼠。②实验开始前,人参二醇皂甙3个剂量组:分别舌下静脉注射22.5,45.0,90.0mg/kg剂量的人参二醇皂甙(由吉林大学基础医学院天然药物研究室提供,将人参加水煮三四次,过滤,水洗脱致无色,再用乙醇洗脱,回收乙醇得浸膏,用乙醇静止2h,回收乙醇得人参二醇皂甙)注射液2.5mL/kg;地塞米松组:舌下静脉注射2mg/kg剂量地塞米松(天津药业集团新郑股份有限公司提供,1mL/支,国药准字H14022804)注射液2.5mL/kg;模型组注射等容量的50g/L葡萄糖注射液。10min后各组分别舌下静脉注射5mg/kg内毒素,建立内毒素休克模型。血压下降至实验前基础血压的2/3时判定为休克状态。③按购自南京建成生物工程公司的相应试剂盒操作,采用722S型分光光度计测定休克后120和240min大鼠肺组织超氧化物歧化酶活力和丙二醛含量。④组间计量资料比较采用t检验。结果:大鼠100只均进入结果分析。①地塞米松组和人参二醇皂甙中剂量组大鼠休克后240min肺组织超氧化物歧化酶活性明显高于模型组(P<0.05),丙二醛含量明显低于模型组(P<0.05)。②人参二醇皂甙各剂量组和地塞米松组大鼠休克后120min肺组织超氧化物歧化酶活力高于模型组,但差异不明显(P>0.05)。结论:人参二醇皂甙中剂量(45mg/kg)能提高内毒素诱导休克后240min大鼠肺组织抗氧化能力,对肺组织有保护作用,该作用与地塞米松相当。AIM: To investigate the protective effect of panaxadiol saponin (PDS) on lung tissue of endotoxin-induced shock rat models. METHODS: The experiment was completed at Research Center of Functional Experimental Sciences of Basic Medicine College of Jilin University from May to December 2003. ① Totally 100 healthy Wistar rats of either gender aged 2.5-3 montbs were divided into 5 groups according to random lot method: model group, dexamethasone (DEX) group, low-, middle- and high-dosage PDS groups. And then 10 rats of each group were killed 120 minutes and 240 minutes after shock. ② Before experiments, rats in 3 PDS groups were injected with 2.5 ml,/kg PDS of dosage of 22,5, 45.0 and 90,0 mg/kg into vein under lingua, respectively. PDS which was provided by Pharmacognosy Department of Jilin University was isolated by 3-4 times of water extract, filtration, elution, reclaim and silence with ethanol. The rats in DEX groups were injected with 2,5 mL/kg DEX of dosage of 2 mg/kg into vein under lingua. DEX was provided by Tianjin Pharmaceutical Company (1 mL/tube, H14022804). The rats in model groups were injected with the same volume of glncose (50 g/L). 5 mg/kg endotoxin was injected into vein under ligua after 10 minutes establish endotoxin-induced shock models. Shock state was judged by declining to 2/3 of basal blood pressure. .③ The activity of superoxide dismutase (SOD) and level of malondialdehyde (MDA) in lung tissue were assayed 120 minutes and 240 minutes after shock with 722S spectrophotometer according to the kits of Jiancheng Bioengineering Company. ④ t test was adopted by comparing data between groups. RESULTS: Totally 100 rats entered the final analysis, ① SOD activity in lung tissue was higher in DEX group and middle-dosage PDS group than that in model group 240 minutes after shock (P 〈 0.05), but MDA content was lower in above two groups than that in model group (P 〈 0.05). ② SOD activities in lung tissue were higher in PDS dosage groups and DEX
