摘  要:从奥利亚罗非鱼精巢提取总RNA,采用RT和3’RACE(rapid amplification of cDNA ends)法分离和测定了奥利亚罗非鱼DMRT1(DM-related transcription factor1)cDNA的3’序列,扩增得到1086bp的片段,阅读框共编码254个氨基酸,序列分析结果表明,奥利亚罗非鱼DMRT1基因的氨基酸序列与尼罗罗非鱼DMRT1基因的同源性为99%,与黑鲷、黄鳝、新月鱼、虹鳟、青鳉鱼等动物的DMRT1cDNA和尼罗罗非鱼、奥利亚罗非鱼等动物的DMO(DM-domain gene in Ovary)cDNA的同源性分别为:80%、78%、73%、62%、41%、26%、26%。说明DM-RT1基因具有高度的保守性,在不同物种中差别较小,但是与DMO的同源性很低。Total RNA was isolated from testis of Oreochromis aurea using high pure RNA isolation kit. The cDNA of DMrt1 (DM-related transcription factor1)was amplified by 3' RACE PCR(Rapid Amplification of cDNA Ends -Polymerase chain reaction)strategy using the total isolated RNA as template. We amplified a fragment about 1086bp, encoding 254 amino acids. Together with 3' end noncoding regions, sequence analysis revealed homology of DMrtl cDNA of Oreochromis aurea and Oreochromis niloticus was 99%. We compared the alignment of deduced amino acid between DMRT1 cDNA from Acanthopagrus schlegelii , Monopterus albus , Xiphophorus maculatus ,Oncorhynchus mykiss , Oryzias latipes , and DMO (DM-domain gene in Ovary) cDNA from Oreochromis aurea, Oreochromis niloticus. The homology was 80%, 78%, 73%, 62%, 41% , 26%, 26% respectively. These results indicated that DMRT1 cDNAis highly conservative and there is great difference in sex-determining mechanism of milter and spawner.
