良种沙棘“实优1号”组织培养研究

摘　　要：以良种沙棘“实优1号”的水培茎尖和叶片为外植体进行了不定芽及愈伤组织诱导试验。结果表明,水培茎尖在1/2M S+6-BA 1.0 m g.L-1+IAA 0.5 m g.L-1上可诱导出大量愈伤组织及不定芽,继代转接可获得丛生芽。幼苗茎段在1/2 M S+6-BA 2.5 m g.-L 1+IAA 0.2 m g.L-1上可诱导腋芽增殖,但倍数较低;水培叶在1/2 M S+6-BA 0.5 m g.L-1+KT 0.2 m g.L-1+NAA 0.02 m g.L-1上诱导效果好,不定芽多,分化速度快,30 d平均分化不定芽9.1个,分化率达94.1%。继代苗在1/2 B5+6-BA 0.3 m g.L-1+IBA 0.4 m g.L-1上有较高的生根率,达到87.5%。The hydroponics stem apexes and leaves of ＂Shiyou 1＂（Hippophae rhamnoides ） were used as explants to induce adventitious buds and callus. The results showed that adventitious buds and callus could be obtained from stem apexes on 1/2 MS ＋ 6-BA 1.0 mg ·L^-1＋ IAA 0. 5 mg ·L^-1, and cluster shoots were got by subculture. Axillary buds could be induced on 1/2 MS＋ 6-BA 2. 5 mg ·L^-1＋ IAA 0.2 mg ·L^-1 from juvenile stems, but the propagation rate was low. On 1/2 MS ＋ 6-BA 0. 5 mg ·L^-1＋ KT 0.2 mg·L^-1＋ NAA 0. 02 mg ·L^-1, an average of 9.1 adventitious buds per explant were induced effectively and easily from hydroponics leaves after thirty days, the differentiation rate was up to 94. 1%. On 1/2 B5 ＋ 6-BA 0. 3 mg ·L^-1＋ IBA 0.4 mg ·L^-1, roots were induced with a high inducing rate of 87, 5% on the subcuhured plantlets.
