摘 要:目的:建立反相高效液相色谱法测定大豆苷元-3′-磺酸钠及其胶囊的含量。方法:采用 Hypersil ODS(250 mm×4.0 mm,5μm)色谱柱,甲醇-10 mmol·L^(-1)磷酸二氢钾溶液(35:65)为流动相,对羟基苯甲醛为内标物,流速0.8 mL·min^(-1),检测波长为254 nm,柱温:30℃。结果:大豆苷元-3′-磺酸钠在1~40μg·mL^(-1)范围内峰面积与其浓度呈良好的线性关系(r=0.9999);样品平均回收率为99.49%。结论:该方法专属性强,结果准确,重现性好,可用于大豆苷元-3′-磺酸钠制剂含量的质量控制。Objective: The RP - HPLC assay was developed to establish method to determine the content of sodium daidzein - 3' - sulfonate (SDS). Methods: The method was performed on a column of Hypersil ODS (250 mm×4.0 mm, 5 μm), using MeOH-10 mmol. L^-1 KH2PO4(35:65)as the mobile phase at the detective wavelength of 254 nm with the flow rate of 0.8 mL·min^-1. p-Hydroylbenzaldehyde was employed as the internal standard. Results: Satisfactory separation among SDS and related impurities was obtained. The calibration curve was linear overthe range of 1 - 40 μg·mL^-1 for SDS(r = 0.9999). The average recovery of SDS was 99.49%. Conclusion: The method is simple, sensitive and accurate and can be used for the quality control of SDS. The proposed method has been applied to the determination of SDS in medicine with satisfactory results.
