摘  要:用淀粉酶和蛋白酶除去蔗渣和麦草样品中的淀粉和蛋白质,然后用不同的温度、时间、NaOH浓度和料液比对经过预处理的原料分离半纤维素。对得到的半纤维素进行相对纯度和得率分析,算出聚戊糖得率,并用FTIR(傅立叶变换红外光谱)分析了分离的半纤维素结构,探讨了分离条件与得率以及相对纯度的关系。实验结果为:蔗渣原料最佳抽提条件为碱浓8%、抽提温度45℃、时间20h、料液比1∶20;麦草原料最佳抽提条件为碱浓4%、温度45℃、时间4h、料液比1∶20。Amylase and proteinase were used for eliminating the amylum and protein in bagasse and wheat straw sample. Then different temperature, time, NaOH concentration and liquor ratio were adopted to separate hemicellulose from the materials mentioned above. The yield and purity of hemicelluloses were analyzed and the yield of pentosan was calculated, and the structure of the hemicellulose was also analyzed by FTIR( Fourier transform infra- red), then the relation between separating condition and yield and purity of the hemicellulose was analyzed. The results of this experiment indicate that for bagasse, the optimal extraction conditions are: NaOH concentration 8%, temperature 45℃, time 20h, solid-to-liquid ratio 1 : 20 ; for wheat straw, the optimal extraction conditions are : NaOH concentration 4%, temperature 45℃ time 4h, solid-to-liquid ratio 1 : 20.
