摘  要:目的以RP-HPLC测定人血浆中盐酸二甲双胍的浓度。方法色谱柱为Diamosil C18(5μm,4.6mm×250mm),流动相为0.01mol·L^-1 KH2PO4 -006mol·L^-1 SDS-乙腈(34.25:34.25:31.5),流速为1.0ml·min^01,检测波长为233nm,内标为阿替洛尔。结果盐酸二甲双胍的线性范围为0.05-4.00μg·ml^-1,回归方程为Y=1.3397X+0.0222(r=0.9999);日内精密度为1.25%~6.14%,日间精密度为0.93%-6.43%。结论本方法操作简便,测定结果准确,重复性好,可用于盐酸二甲双胍人体药代动力学研究。Objective To establish a RP-HPLC for the determination of Metformin Hydrochloride in human plasma. Method The Diamosil C18 column (5 μm, 4.6 mm ×250 mm) was used with mobile phase of 0.01 mol·L^-1 KH2PO4 -0.006 mol· L^-1 SDS-acetonitrile (34.25: 34.25: 31.5). The flow-rate was 1. 0 ml ·min^-1. The detection wavelength was 233 nm. Internal standard was Atenolol. Result The linear range of the calibration was (0.05 - 4, 00) μg·ml^-1 ( Y = 1. 3397X + 0. 0222, r = 0, 9999). Within-day RSD was 1. 25% -6.14%. Between-day RSD was 0.93% % - 6.43%. Conclusion The method is simple, accurate and was used in the phamarmacokinetic study of Mettformin Hydrochloride.
