肾综合征出血热患者T淋巴细胞诱导凋亡的研究

摘  要:目的研究肾综合征出血热(HFRS)患者外周血T淋巴细胞(PBL-T)激活诱导细胞死亡(AICD)的发生情况,探讨AICD在肾综合征出血热发病机理中的作用。方法以免疫磁珠负性分离PBL-T;应用植物血凝素(PHA)和离子霉素(ionomycin)体外培养诱导细胞活化;采用HE染色和TUNEL染色观察凋亡情况并用流式细胞仪检测PBL-T的凋亡率和淋巴细胞亚群分布。结果采用免疫磁珠分离的PBL-T体外培养12 h后,经诱导发生凋亡的细胞出现DNA断裂;HFRS患者诱导组PBL-T凋亡率为(27.79±0.99)%,明显高于未诱导组(17.16±1.14)%,P<0.01;正常人PBL-T诱导与否其凋亡率差异无显著性(11.29±0.66)%(、10.94±0.71)%(P>0.05),无论是否诱导,患者PBL-T的凋亡率均高于正常人。结论以植物血凝素和离子霉素可有效地诱导体外培养PBL-T的凋亡;HFRS患者PBL-T经激活诱导后凋亡率明显提高;无论诱导与否患者PBL-T的凋亡率均高于正常人;HFRS患者PBL-T存在AICD现象,并且与EHFV感染有关。OBJECTIVE To study the activation-induced cell death (AICD) in peripheral blood T-lymphocytes (PBL-T) from patients with hemorrhagic fever with renal syndrome (HFRS), and the effect of AICD in the pathogenesis of HFRS. METHODS The PBL-T of 14 patients were isolated with negative selection by magnetic beads, and cultured with or without PHA and ionomycin. The apoptotic cell death of PBL-T was detected by TUNEL staining and assessed by flow cytometry. RESULTS In HFRS patients, the apoptotic rate of PBL-T activated with PHA and ionomycin was significantly higher than that of without treatment [(27.79 ± 0. 99)% vs (17.16±1.14)%, P〈0.01]; there was a similar apoptotic rate in healthy control between activation and unactivation [(11.29±0.66)% vs 10.94±0.71)%, P〉0.05]; of all, the ratio of apoptotic cell death of PBL-T in HFRS patients was higher than that of healthy control(P〈0.01). CONCLUSIONS Activation-induced cell death in peripheral T cells exists in patients with HFRS, and the AICD of T lymphocytes may play an important role in the pathogenesis.
