补脾方药对脾虚大鼠壁细胞胃泌素受体及细胞内[Ca^(2+)]i作用的观察

摘　　要：目的:观察脾虚模型大鼠壁细胞胃泌素受体及受体后信号细胞内钙离子的变化,并观察补脾方药党参、白术、补中益气汤对其的影响。方法:大鼠壁细胞采用EDTA-Pronase酶法消化而来。胃泌素受体检测采用放射配基结合实验,壁细胞内[Ca2+]i变化测定采用双波长荧光法。结果:大黄脾虚模型大鼠壁细胞胃泌素受体结合位点数有显著降低,经补脾方药治疗后有所恢复;同时脾虚模型壁细胞内静息钙离子浓度明显低于正常组,经胃泌素刺激后壁细胞内[Ca2+]i有所升高,且脾虚模型升高幅度明显高于正常组,经治疗后此状态有所恢复,但无统计学意义。结论:脾虚证可能与胃泌素受体低表达相关,且壁细胞内钙离子处于低代谢状态,但对于胃泌素刺激的反应性增强,进而可能导致酸分泌敏感性相对增高,胃黏膜易损性增强。补脾方药对其有一定的调节作用。Objective ： To observe the changes of gastrin receptor and intracellular calcium level of parietal cells in spleen-deficiency rats and che regulating effects of herbs or recipe for invigorating spleen such as Dangshen, Baizhu, Buzhong Yiqi decoction. Methods：the parietal cells isolated from rats were made by EDTA- Pronase method. Radioligand binding assay was established in screening gastrin receptor of parietal cells. [Ca^2＋]i was measured Using Fura-2/AM fluorescence method. Results：the gastrin receptor binding sites of parietal cells in spleen-deficiency rats were obviously lower than that in the normal group, and binding sites were increased remarkably by administration of Baizhu and Buzhong Yiqi decoction. The resting [ Ca^2＋ ]i of parietal cells in spleen-deficiency rats were significantly lower than that of the normal rats. But the elevating percentage of [ Ca^2＋]i stimulated by gastrin were higher markedly than that of the normal group. The herbs or recipe for invigorating spleen had some effects of regulating the disorder conditions. Conclusions：there is a close relationship between the spleen-deficiency syndrome and the lower expression of gastrin receptor of parietal cells.The resting [Ca^2＋ ]i of parietal cells was in a lower metabolic condition in spleen-deficiency syndrome, but the response of parietal cells increased after stimulated by gastrin, which may lead to the sensitivity of gastric acid secretion increased and the vulnerability of gastric mucosa. The herbs or recipes of invigorating spleen might have some regulating effects.
