摘 要:目的:探讨姜黄素对结肠癌细胞抗失巢凋亡的影响。方法:采用不同浓度的姜黄素处理结肠癌SW-480细胞,采用末端脱氧核苷酸转移酶介导的dUTP切口末端标记法(TUNEL)法检测结肠癌细胞抗失巢凋亡,采用软琼脂集落形成试验检测细胞的生长增殖。结果:姜黄素能有效地抑制结肠癌SW-480细胞锚着不依赖性增殖,且呈浓度依赖性。结肠癌细胞经姜黄素处理后,可促进失巢凋亡且与时间和浓度相关。结论:姜黄素可有效地抑制结肠癌细胞锚着不依赖性增殖,其机制可能与促进失巢凋亡有关。Objective : To study the effects of curcumin on anoikis resistance in human colon cancer cells. Methods: After colon cancer SW-480 ceils were treated with curcumin, the growth of anchorage independence of colon cancer SW-480 cells was examined in soft agar colony formation, and apoptosis of colon cancer were evaluated by terminal deoxynucleatidyl transferase-mediated deoxyuridine triphosphate nick end labeling (TUNEL) and DNA ladder. Results: Curcumin could significantly inhibit the growth of anchorage independence of colon cancer SW-480 cells in soft agar colon formation in a dose-dependent. Curcumin could induce apoptosis of colon cancer cells from DNA ladder and TUNEL assay, which were in a time-and dosedependent manner. Conclusions: Curcumin could inhibit the growth of anchorage independence of colon cancer SW-480 cells, which was related to anoikis.
