摘  要:目的观察附加二氮嗪(DZ)与1,6-二磷酸果糖(FDP)心麻痹液对冷保存大鼠心肌能量代谢的作用。方法42只SD大鼠随机分为4组,正常对照组6只;其余各组每组12只,分别为STH组(单纯St.Thom asⅡ号液),DF组(St.Thom asⅡ号液+DZ 50μmol/L+FDP 5 mmol/L),DF+5-HD组(St.Thom asⅡ号液+DZ50μmol/L+FDP5mmol/L+5-羟基葵酸盐100μmol/L),在4℃条件下保存心脏6 h后,6只取左心室,测定心肌ATP、ADP、AMP含量,计算能荷;提取线粒体,测定线粒体内游离Ca2+浓度([Ca2+]m)及线粒体呼吸功能;另外6只建立离体左心工作模型,再灌注30 m in后,测量上述指标。结果与正常对照组比较,冷保存后STH组心肌ATP含量及能荷显著下降,[Ca2+]m显著增高,线粒体Ⅲ态呼吸速率(ST3)及呼吸控制比(RCR)明显降低(P<0.01);再灌注后心肌ATP含量及能荷没有明显改善,[Ca2+]m进一步增高,RCR进一步降低(P<0.01)。DF组各项指标显著优于STH组(P<0.01);线粒体ATP敏感性钾通道阻断剂5-HD可部分降低附加DZ与FDP的保护效果。结论附加DZ与FDP可显著改善St.Thom as液冷保存大鼠心脏的能量代谢。Objective To evaluate the effect of cardioplegia solution supplemented with diazoxide (DZ) and fructose-1,6-diphosphate (FDP) on myocardial energy metabolism in preserved rat hearts. Methods Forty-two SD rats were divided into 4 groups randomly including the control group, STH group ( St. Thomas Ⅱ solution), DF group (St. Thomas Ⅱ solution with DZ 50 μmol/L + FDP 5.0 mmol/L) and DF +5-HD group ( St. Thomas Ⅱ solution with DZ 50 μmol/L + FDP 5.0 mmol/L + 5-hydroxydecanoate 100 μmol/L), with 6 rats in the control group, and 12 each in the other groups. Except the control group, 6 hearts were stored at 4 ℃ for 6 h, and another 6 hearts were reperfused for 30 min after stored at 4 ℃ for 6 h. The content of myocardial ATP, ADP, AMP were determined and energy charge (EC) were calculated. The mitochondrial Ca^2+ concentration ( [ Ca^2+ ] m ) and respiration function were also determined. Results As compared with normal controls, after stored at 4 ℃ for 6 h or reperfused for 30 min, the content of myocardial ATP, EC and mitochondrial respiratory control rate (RCR) decreased, and [ Ca^2+ ] m increased significantly ( P 〈 0.01 ) in STH group. DF group had better results than STH group. Mitochondial ATP-sensitive potassium channel blocker 5-HD could partially decrease the effect of diazoxide and fructose-1,6-diphosphate on myocardial energy metabolism. Conclusion The results suggested that St. Thomas solution supplemented with diazoxide and fructose-1,6-diphosphate improve myocardial energy metabolism in hypothermic preserved rat hearts.
