摘  要:目的 建立同时测定加味左金丸中盐酸小檗碱和吴茱萸次碱含量的反相离子对液相色谱法.方法 采用Agilent ZORBAX SB-C18柱(4.6 mm×250 mm,5 μm),甲醇-乙腈-15 mmol·L^-1 SDS水溶液(20:40:40,pH 7.5)流动相,检测波长343 nm.结果 盐酸小檗碱和吴茱萸次碱的线性范围分别为12~120和0.45~6.81 mg·L^-1,盐酸小檗碱的回收率为99.21%,RSD=1.60%;吴茱萸次碱的回收率为101.40%,RSD=3.32%.结论 采用本方法同时测定加味左金丸中盐酸小檗碱和吴茱萸次碱含量具有简便、快速、准确等优点.OBJECTIVE To develop a new method for the simultaneous quantitative determination of berbefine hydrochlofide and rutcerpine in Jiawei Zuojin Pill by reversed phase high performance liquid chromatographic method. METHODS The optimum chromatographic conditions were as follows: Agilent Zorbax SB - C18 column(4.6 mm×250 mm,5 μm), methanol-acetonitrile-H2O( containing 15 mmol· L^-1 SDS) (20:40:40,pH 7.5) and the detection wavelength of 343 nm. RESULTS The calibration curves were linear with the range of 12 - 120 mg·L^-1 for berberine hydrochloride ( n = 5, r = 0.999 5) and 0.447 - 6.81 mg·L^-1 for rutaecarpine( n = 5, r = 0.999 6). The recovery and RSD of berberine hydrochloride were 99.21% and 1.60% and those of rutaecarpine were 101.40% and 3.32%. CONCLUSION This method is successfully applied to determine the contents of berberine hydrochloride and rutaecarpine in Jiawei Zuojin Pill.
