甘肃3个特有少数民族回族和汉族DYS287 DYS19的遗传多态性研究

摘　　要：以中国甘肃的3个特有少数民族,天水汉族、天水回族、临夏回族6人群为研究对象,采用 RCR 技术及变性聚丙烯酰胺凝胶电泳法,分析了高变染色体特异的微卫星 DYS19的等位基因分布规律,共检出7种等位基因．甘肃3个特有少数民族 DYS19的等位基因频率存在显著性差异(P＜0．05)．通过 PCR 方法调查了 Alu 元件插入基因座 DYS287的多态性．结果显示,除保安族没有发现 Alu 插入外,其他人群均发现 Alu 插入,其中临夏回族表现了较高的插入频率(12．9％)．MDS 分析表明：裕固族和汉族的遗传关系比较接近,6人群遗传距离和空间距离没有明显的相关性．Y-chromosome specific microsatellite DYS19 and DYS287 were genotyped in 400 males of six populations in Gansu by using the method of PCR followed by denaturing polyacrylamide gel electrophoresis and silver staining. The six populations are Dongxiang, Yugur, Bonan and three specific ethnic minority groups： Tianshui Han, Linxia Hui and Tianshui Hui in Gansu. The analysis of the DYS19 revealed a high variability： seven alleles （182 bp, 186 bp, 190 bp, 194 bp, 198 bp, 202 bp, 206 bp） have been found and the frequency ranges from 0.0128 to 0.654. The pairwise comparison with Fisher＇s exact test revealed that there existed significant difference （P 〈 0.05） in DYS19 phenotype distribution among three specific ethnic minority groups in Gansu（Bonan, Yugur, Dongxiang）. The polymorphism of the Y-chromosomspecific Alu insert sequence DYS287 was also investigated in six Gansu populations. The result showed that Alu element was absent in Bonan, and presented in all the other populations. The highest frequency of the insertion was found in Linxia Hui （12.9%）. The MDS of parewise Fst proved that genetic relationship of all the populations did not present apparent geographical difference.
