摘 要:目的探讨摸索一种简便可行,用来提取肺脏线粒体的方法。方法利用不同分离介质,通过低温离心机在不同转速下分离出肺脏线粒体,用固定液固定,在电镜下观察。结果两种方法都能分离出肺脏线粒体,但以氯化钾溶液作为分离介质提取的肺脏线粒体有肿胀、自溶和破碎,而以STE液为分离介质的方法能够分离出完整的肺脏线粒体。结论以STE液为线粒体分离介质来提取肺脏线粒体的方法是一种简便可行的实用方法,可用来观察药物对急性肺损伤时线粒体影响的研究。Objective To establish a simple and feasible method for isolation of mitochondria from lung in rat. Methods Using different isolation mediums, rat pulmonary mitochondrial pellet was collected by differential centrifugation. Then accumulated mitoehondria was fixed with fixing solution. Lastly, mitochondrial ultrastrueture was observed by transmission electronic microscope. Results To compare the results of different separating mediums used to isolate mitoehondria of rat lung, on the electron mierographs, pulmonary mitochondria isolated in kalium chloratum were swollen, autolytie or broken, but the majority of mitochondria in rat lung separated by STE solution were intact. Conclusion The method of STE isolation medium is simple and reliable. Mitochondria can be totally accumulated in this way.
