摘  要:目的观察FALL-39和头孢他定(CFZ)治疗脓毒血症的疗效。方法从pGEX-1λT-FALL-39转染的E.coli JM109中提纯FALL-39,检测FALL-39对绿脓杆菌(PAG)的杀菌活性。采用更生霉素(AMD)增强BALB/c小鼠对PAG感染的敏感性,建立脓毒血症模型,观察FALL-39和CFZ治疗脓毒血症的疗效。结果FALL-39分子量约5kDa,纯化量约600-650μg,对PAG的最低抑菌浓度(MIC)、最低有效浓度(MEC)、最低杀菌浓度(MBC)分别为25、100、50μg/ml。盐水组、脓毒血症组、CFZ治疗组、FALL-39治疗组和CFZ+FALL-39治疗组小鼠死亡率分别为0%、75%、62.5%、37.5%和12.5%。结论FALL-39治疗脓毒血症比CFZ更为有效,两者联用效果则更佳。Objective To investigate the effects of FALL-39 peptide and cefatazigine on treating sepsis. Methods Purified FALL-39 was extracted from Escherichia coli JM109 transfected by the pGEX-1λT-FALL-39 recombinant vector. The antibacterial activity of FALL-39 was verfied by measuring the MIC, MEC, MBC for P. aeruginosa. The infectional sensitivity of BALB/c mice for P. aeruginosa was induced by employing actinomycin D and the actinomycin-sensitized BALB/C murine sepsis model was constructed. The treatment effects of FALL-39 peptide and cefatazigine on BALB/C murine sepsis model were investigated. Results The molecular magnitude of FALL-39 was identified approximate 5kDa. About 600- 650μg of highly purified FALL-39 was obtained. The antibacterial activity of FALL-39 was determined,and its minimal effective concentration(MEC),minimal inhibitory concentration(MIC), and minimal bactericidal concentration(MBC) were 25,100, and 50μg/ml, respectively. Death rates of saline, sepsis, CFZ, FALL-39 recombinant peptide and CFZ+FALL-39 subgroups were 0%,75%,62. 5%,37.5%,and 12.5%,respectively. Conclusion FALL-39 recombinant peptide is more effective than ceftazigine in treating sepsis and the efficacy of combining ceftazigine with FALL-39 recombinant peptide in treating sepsis is more than FALL-39 or ceftazigine alone.
