摘  要:目的:观察抗氧化剂丙丁酚对STZ导致的糖尿病小鼠胰岛β细胞功能的影响。方法:分正常对照组,正常对照治疗组,糖尿病组,糖尿病治疗组。用1%的丙丁酚加入标准的小鼠饲料中,进行预防性干预,小鼠腹腔注射STZ后在不同时间测空腹血糖、血浆胰岛素,做腹腔葡萄糖耐量试验,取胰腺标本作病理切片。结果:丙丁酚干预组0w、1w、2w、3w小鼠空腹血糖均较糖尿病组明显降低,且有显著性差异;3w时丙丁酚干预组腹腔葡萄糖耐量试验显示空腹及葡萄糖注射后0.5h、1h、2h其血糖均较糖尿病组降低,且有显著性差异(P<0.05或0.01)。3w时糖尿病预防给药组空腹血清胰岛素水平较糖尿病组明显升高(P<0.01)。结论:抗氧化剂丙丁酚有降低STZ导致的糖尿病小鼠的空腹血糖及改善糖耐量的作用;抗氧化剂丙丁酚通过保护STZ导致的糖尿病小鼠胰岛β细胞功能而降低血糖的作用。Objective: To study the effect of probucol on pancreatic β -cells in STZ-induced diabetes mice. Methods: The mice were randomly divided into four groups with ten mice in each group. The first group was given standard food (C group), The second group given standard food and 1% probucol (C + T group). The third group given standard food and treated with multiple low-dose of STZ (D group), and the forth group given standard food, 1% probucol as well as treated with multiple low-dose of STZ (D + T group). The preventive intervention was undertaken with standard diet mixed with 1% probucol in Kunming mice. At different time after STZ was injected intraperitoneally, fasting blood glucose was measured. Serum insulin and glucose tolerance test was done. The pancreatic pathological cbanges were investigated under hght microscope. Results: The fasting blood glucose at 0,1,2,3 weeks in D+C group was lower than that in D group (P 〈 0.05 or P〈 0.01). At 3 weeks in D+C group glucose tolerance was significantly improved (P〈 0.05 or 0.01) and compared with the D group, serum fasting insulin in D+C group was increased (P〈0.01). Conclusion: Probucol could reduce fasting blood glucose and improve glucose tolerance by protection of pancreatic β -cells in STZ-induced diabetes mice.
