摘 要:目的研究中国汉族人群人类血小板抗原HPA15(Gov)系统多态性,评估其在血小板配型输注中的作用。方法采用PCRSSP技术对1000名来自不同省份汉族无关献血者进行HPA15系统基因分型。结果HPA15在所调查的7个省份汉族人群中的分布差异无显著性意义,在1000名汉族中HPA15a和15b基因频率分别为0.5320和0.4680,与HardyWeinberg平衡吻合;与其它种族的HPA15分布相比较,也无显著性差异。在随机输血中HPA15a和15b抗原不配合的机会分别为0.1711和0.2029。结论本调查数据表明,随机输血中供受者HPA15抗原不配合比例在中国汉族人群中高达37%,这对研究同种免疫血小板减少症和开展血小板同型输注具有指导意义。Objective To study the polymorphism of the human platelet antigen HPA-15 (Gov) system in Chinese Han population, and evaluate its role in matched platelet transfusion. Methods A total of 1,000 unrelated Chinese Han blood donors from different provinces of China were genotyped for HPA-15 system using PCR-SSP assay. Re- suits The distributions of the HPA-15 alleles were no significantly difference among the Chinese in the investigated provinces. The distributions of HPA-15 in 1,000 Chinese fit Hardy-Weinberg equilibrium. Gene frequencies of HPA- 15a and HPA-15b were 0. 5320 and 0. 4680, respectively. In comparison to other ethnic groups, no significant differ- ences were observed in the distributions of HPA-15. The chances of incompatibility in random platelet transfusion were 0. 1711 and 0. 2029 for HPA-15a and 15b antigens, respectively. Conclusion The distributions of HPA-15 sys- tem in Chinese Han people were investigated in such a large scale for the first time. The present data indicate that the chance of HPA-15 antigen incompatibility in random platelet transfusion is up to 37 % and provide a basis for further study on HPA-matched transfusion.
