摘 要:从云南省西双版纳傣族自治州3家医院采集到发热病人血清106份,脑炎病人脑脊液5份,从中分离到27株病毒。经血清学鉴定,该类病毒与披膜病毒科、黄病毒科、布尼亚病毒科某些病毒的免疫腹水,以及呼肠孤病毒科环病毒属M14、YN187病毒的免疫腹水,都不发生反应。电镜观察病毒为球形无囊膜颗粒,直径65.49±3.11nm,其核衣壳直径为51.79±2.01nm。在感染细胞中常见到大量颗粒状包涵体。负染中可见排列整齐的壳粒结构,显示了典型的环状病毒形态特征。该类病毒对酸和热敏感,抵抗5—Id?和乙醚。27株病毒的基因组均由12条RNA片段组成,其带形均相同,分子量约为0.25×10~6~2.5×10~6道尔顿。该类病毒的带形与文献报道的任何一种环状病毒的RNA带形都不同。病人双份血清中对该环状病毒特异IgM抗体滴度显示4倍以上升高。对该类病毒在分类学上的地位和对无名热和脑炎的病原学意义进行了讨论,目前暂定名为BANNA病毒。In the summer of 1987, 25 strains of virus were isolated from 106 sera of patients with unknown fever and 2 strains from 5 cerebrospinal fluids of patients with encephalitis collected in Dai Nationality Autonomous Prefecture of Xi Shuang Ban Na of Yunnan ( BANNA ). By EIA all the 27 isolates were serologically unrelated to any of the 29 reference antibodies used in the study. The electron micrographs of ultrathin section of C6/36 cells infected with the isolates revealed virions morphologically similar to orbivirus, spherical, without envelope, 65.49±3.11 nm in diameter, with double-layered capsid. The virions were frequently observed in the cytoplasm and associated with granular inclusion bodies. Negative-contrast examination showed virions 68.30±3.23 nm in diameter, nucleocapsids 55.04 ±2.35 nm in diameter, with icosahedral symmetry and clearly discernible capsomeric structures characteristic of orbivirus. The virus was resistant to ether and 5-IdU, and unstable at 56℃ for 30 minutes and below pH 3.0 at 37℃ for 2 hours.The virus genomes were found to contain 12RNAseg-'ments, with molecular weight ranging from 0.25×108 to 2.5×106 dalton. The profiles of genome RNA shared by all the 27 isolates were different markedly from those of the published orbiviruses. It was suggested that the BANNA orbivirus is a new member of the genus Orbivirus of Reoviri-dae. The etiological relationship of BANNA orbivirus with unknown fever and encephalitis is discussed.
