海南省两株环状病毒的分离与鉴定

摘　　要：环状病毒属(Orbivirus genus)由许多病毒所组成,其中一些可在脊椎动物和昆虫体内增殖并通过吸血昆虫传播,属呼肠孤病毒科的虫媒病毒。有些环状病毒可引起家畜发病,如蓝舌病毒(BIV)、鹿流行性出血热病毒(EHDV)和非洲马瘟病毒(AHSV)等。而环状病毒中的科罗拉多蜱热病毒(CTFV)和科麦罗沃病毒(KEMV)与人类疾病有关,其传播媒介均为蜱,主要症状为发热,寒颤,头痛和背、腿部肌肉剧痛。因而国际上对这些环状病毒的研究愈来愈深入。我国在这方面起步较晚,1981年梁雪嘉等从北京郊区捕获的三带喙库蚊中分离到一株对乳鼠非致病的环状病毒(M14);1985年7月徐普庭等从云南省的中华按蚊和棕头库蚊体内分离到两株对3周鼠致死的环状病毒。1985年5月,我们从海南省三带喙库故中分离到HN131株,从麻翅库蚊分离到HN295株,以及从其它蚊子中分离到4株病毒,经电镜和核酸电泳初步鉴定为环状病毒。现将前两株病毒的鉴定结果报告如下。During the summer of 1985, 2 strains of orbivirus ( HN131 and HN295 ) were isolated from Culex bitaeniorhynchus and Culex tritacniorhynchus collected in Hainan Province. Electron microscopic examination of cell cultures infected with virus ( HN295 ) revealed the morphological features of orbivi-rus2 spherical particles, 56.98±2.54nm in diameter and a diffuse, structureless outer capsid layer. The results of serological identification by enzyme immunoassay ( EIA ) on virus-infected cell cultures showed that the two strains of virus were antigenically not related to Alphaviruses, Flaviviruses and Bunyaviruses. Neutralization test revealed that the two strains of virus were identical. The isolates were resistant to ether and 5-Brdur, labile at low pH values and 56℃. The genome of virus ( HN295 ) contained 12-seg-mented RNA. The profile of viral RNA PAGE ( HN295 ) was identical to that of viruses ( HN131 and BNC 626). The molecular weights of RNA segments varied from 0.325 to 1.83×106 daltons and the total molecular weight was about 10.175×106 daltons.
