摘 要:将中华绒螯蟹(Eriocheir sinensis)血清凝集素与动物(鳖、鲫鱼、鸡、鸽、兔、鼠、人A、B、O型)红细胞和细菌(八叠球菌、嗜水气单胞菌、短小芽孢杆菌、大肠杆菌、葡萄球菌、假单胞菌和拟态弧菌)进行凝集反应.结果表明,中华绒螯蟹血清凝集素有明显的选择性,与小白鼠红细胞和八叠球菌的凝集活性最高(P<0.01).血清凝集价在蟹个体之间变化范围较大,性别之间无显著差异(P>0.05).10 mmol/L的CaCl2和300 mmol/L的NaCl不影响凝集素与小鼠红细胞和细菌的凝集活性,600 mmol/L的NaCl可降低其活性.pH 5~9不影响蟹凝集素活性,但400 mmol/L的EDTA在pH 7.0可部分抑制其活性,100mmol/L的EDTA在pH 5.0则可完全抑制其活性.在细菌刺激下,中华绒螯蟹血清凝集素活性在2d达到峰值,随后逐步缓慢下降.接种细菌(八叠球菌)的数量(3.0×109~9.0×109)显著影响血清凝集素的产生(P<0.05),而环境水温度(18~30℃)和蟹体质量(35~80 g)的影响则不显著(P>0.05).[中国水产科学,2006,13(3):365-370]The serum was collected from health Eriocheir sinensis. Firstly, the binding abilities of the agglutinin to erythrocytes of several species of animals and bacteria were tested. The results showed that the agglutinin had different binding abilities to erythrocytes of chicken, pigeon, rabbit, mouse and human (types A, B and O). However, it could not bind to the erythrocytes of C. auratus and T. sinensis. The binding ability to erythrocytes of mouse was stronger than those of other animals, which had significant difference (P〈0.01). In addition the agglutinin could bind bacteria such as M. sarcina and A. hydrophila (P〈0.01), but couldn't bind E. coli , B. pumilus , S. aureus P. putida nor V. minicus. Secondly, the pattern of agglutinin production was examined, as well as the influencing factors on its production. E. sinensis were injected with bacteria (M. sarcina ), and a peak value of serum agglutinin appeared after two days, then it declined gradually to normal level at day 14. This suggests that the agglutinin serving as a nospecific preventive factor can rapidly respond to a heterogenous stimulator and keep only for a short phase in E. sinensis serum. An experiment according to biostatics principle was designed to examine the effects of three factors such as body weight, water temperature and concentration of bacteria on agglutinin production in vivo. The results showed that there was significant difference on agglutinin level produced among the groups injected with various concentrations of bacteria (P 〈 0.05), but no difference among the groups with body weight from 35 g to 80 g and under the conditions of water temperature from 18℃ to 30℃ (P〉0.05). In addition, there was a greater individual diversity among E. sinensis in agglutinin titers binding M. sarcina from 4 to 32, mouse erythrocyte from 128 to 2 048 and human erythrocyte from 8 to 64, but no difference between male and female groups (P 〉0.05).
