N－羟基琥珀酰亚胺－α－萘乙酸酯（SINA）为柱前衍生试剂反相高效液相色谱分离测定肽及其水解产物

摘　　要：以自行合成的丁二酰亚胺羧酸活性酯类试剂Ｎ－羟基琥珀酰亚胺－α－萘乙酸酯为柱前衍生试剂，用反相高效液相色谱分离测定了谷胱甘肽、三甘肽、二甘肽及谷氨酸、胱氨酸、甘氨酸，在Ｃ１８柱上，采用的流动相为含１５ｍｍｏｌ／Ｌ、ｐＨ＝５．４的醋酸－醋酸钠缓冲溶液和３％的乙酸乙酯的甲醇－水（１７／８０，体积比）溶液，１６ｍｉｎ内６种衍生物得到完全分离，当信噪比为３：１时，检测限达ｐｍｏｌ级，重现性良好．Using N-hydroxysuccinimidyl-α-naphthylacetate(SINA,synthesized by ourselves) as pre-column derivating reagent,glutathion,diglxcyl-glycine,glycyl glycine, Glu,(Cys)2 and Gly were separated and determined by reversed phase high performance liquid chromatograph (RP-HPLC). The six derivatives were eluted within 16min with a mobile phase of methanol-acetic ether-water(17/3/80 ,volume ratio)containing 15 mmol/L pH=5.4 acetate buffer on C18 column,At the ratio of signal to noise S/N=3,the detection limit is in lower picornole range,The reproducibility was good.
